一般描述
测试正常大鼠血清的补体活性并证明具有功能性经典和替代性激活途径。 NRS由雄性混合品种大鼠制备。 收集每份血液样品而不使用抗凝剂，凝固后通过离心分离液体部分。 血清通过0.22微米过滤器过滤，aliqoted和冷冻于-80 Ô C.
对随后解冻的样品进行所有补体活性测试以确保报告的功能活性是当样品解冻时客户将获得的功能活性。 如果NRS储存在-70补体活性的几年稳定Ø C或以下持续。 大鼠补体在正常人血清中不如人补体那样稳定，并且如果允许NRS血清保持解冻超过几个小时（即使其保持在冰上），则可观察到活性的显着降解。 另外，在另外的冻融循环之后可能发生显着的活性损失。
物理特性

功能
使用抗体致敏的绵羊红细胞（CompTech＃B200）测试NRS的经典途径溶血活性，并使用兔红细胞（CompTech＃B300）测试NRS的替代途径功能。 每批提供的分析证书给出了血清的分析和特定效价的描述。
测定
经典途径活动的单位是CH50。 一个CH50单位定义为NRS的输入，产生1×10的50％溶解8 在37温育60分钟时EA（CompTech＃B200）ø在1.5的总反应卷C 毫升 GVB ++。
替代途径活动的单位是AP50。 一个AP50单位定义为NRS的输入，产生1.5×10的50％溶解7兔红细胞（二当在37℃温育30分钟后，CompTech＃B300）ø在75μL的总反应卷C GVB ö含有Mg的终-EGTA浓度为13mM。 大鼠替代途径通常比正常人补体标准血清中的活性低约3倍（更少AP50单位/ mL）。
应用
